Нейродегенеративные процессы являются ключевыми в развитии ряда заболеваний — инсульты, болезни Паркинсона и Альцгеймера, эпилепсия, при черепно-мозговой травме. Они включают в себя первичные и вторичные структурно-функциональные изменения нервной ткани, а также гибель нейронов и полное выпадение функций.</p></sec><sec><title>Цель</title><p>Цель. Изучение проявлений гидропической дистрофии и реорганизации глиоцитоархитектоники в процессе формирования темных нейронов в сенсомоторной коре (СМК) большого мозга половозрелых белых крыс после 40-минутной окклюзии общих сонных артерий (ООСА).</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. На белых крысах линии Wistar моделировали 40-минутную ООСА. Головной мозг фиксировали перфузионным способом. Проводили морфометрическую оценку проявлений отека-набухания, цито- и глиоцитоархитектоники СМК в норме (n = 6, контрольная группа), на 1-е (n = 6), 3-и (n = 6) и 7-е сутки (n = 6) после ООСА. Использовали окраску по Нисслю, гематоксилином и эозином, иммуногистохимические реакции NSE, MAP-2, GFAP и AIF1. Определяли относительную площадь зон отека-набухания, численную плотность нормохромных и темных пирамидных нейронов, олигодендроцитов (ОДЦ), микроглиоцитов (МГЦ).</p></sec><sec><title>Результаты</title><p>Результаты. Высокое содержание (20–50 %) темных нейронов после ООСА сопровождалось увеличением в 3.3 раза относительной площади зон отека-набухания (r = 0.82, p = 0.01). Специфические белки (NSE, MAP-2) большей части темных нейронов сохранялись. Общая численная плотность нейронов СМК через 7 сут после ООСА уменьшалась на 26.4 % (слой III, p = 0.001) и 18.5 % (слой V, p = 0.01). Увеличивалось содержание астроцитов, МГЦ и ОДЦ. Пик численной плотности МГЦ отмечен на 1-е сутки, а ОДЦ — на 7-е сутки после острой неполной ишемии (p ≤ 0.001). Выявленные изменения носили диффузно-очаговый характер.</p></sec><sec><title>Заключение</title><p>Заключение. После 40-минутной ООСА в СМК увеличивалось содержание темных нейронов и, как следствие, появлялись признаки гидропической дистрофии. На этом фоне возрастало количество сателлитарных ОДЦ, астроцитов и МГЦ. Вероятно, отек-набухание, активные МГЦ и астроциты предварительно (в 1-е – 3-и сутки) санируют нервную ткань, обеспечивая ее последующее (7-е сутки) структурно-функциональное восстановление с участием ОДЦ.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Neurodegenerative processes are key in the development of a number of diseases — strokes, Parkinson’s and Alzheimer’s diseases, epilepsy, and traumatic brain injury. They include primary and secondary structural and functional changes in the nervous tissue, as well as the death of neurons and complete loss of functions.</p></sec><sec><title>Aim</title><p>Aim. To study the manifestations of hydropic degeneration and reorganization of glio-cytoarchitectonics during the formation of dark neurons in the sensorimotor cortex (SMC) of the cerebrum of mature white rats in 40 min after the common carotid arteries occlusion (CCAO).</p></sec><sec><title>Materials and methods</title><p>Materials and methods. A 40-minute CCAO was simulated in white Wistar rats. The brain was fixed by perfusion method. Morphometric assessment of manifestations of edema — swelling, cyto- and glio-cyto-architectonics of SMC was performed normally (n = 6, control group), on the 1st (n = 6), 3rd (n = 6) and 7th days (n = 6) after CCAO. The Nissl staining, hematoxylin and eosin, immunohistochemical typing NSE, MAP-2, GFAP and AIF1 were applied. The relative area of edema-swelling zones, the numerical density of normochromic and dark pyramidal neurons, oligodendrocytes (OD), m icrogliocytes (MG) were determined.</p></sec><sec><title>Results</title><p>Results. The high content (20–50%) of dark neurons after CCAO was accompanied by a 3.3-fold increase in the relative area of edema-swelling zones (r = 0.82, p = 0.01). Specific proteins (NSE, MAP-2) of most dark neurons were preserved. The total numerical density of SMC neurons decreased by 26.4% (layer III, p = 0.001) and 18.5% (layer V, p = 0.01) after 7 days of CCAO. The content of astrocytes, MG and OD increased. The peak in the number density of MG was observed in the 1st day, and OD — in the 7th day after acute subtotal ischemia (p ≤ 0.001). The revealed changes were of a diffuse-focal nature.</p></sec><sec><title>Conclusion</title><p>Conclusion. After a 40-minute CCAO, the content of dark neurons in SMC increased and, as a result, signs of hydropic degeneration appeared. Against this background, the number of satellite OD, astrocytes and MG increased.
